
Duchenne muscular dystrophy (DMD) is an X-linked disorder caused by the lack of dystrophin by progressive degeneration of the skeletal muscles. Most studies on DMD understandable focus on the muscles, but the dystrophin also expressed in the central nervous system, potentially resulting in cognitive and behavioral changes.
Animal models are used to develop a more comprehensive neuromonitoring protocols and clinical image acquisition procedure. DMD mice have recently developed an animal model that is aligned with progressive muscle wasting seen in DMD. Here, we studied the brains and temporalis muscle structure and neurochemistry of the wild type (WT) and dystrophic (DMD) mice using magnetic resonance imaging and spectroscopy.
Both structural and neurochemical changes observed in the brains of mice DMD and temporalis muscles. There was a decrease in brain volume absolute (WT = 1579 mm3; DMD = 1501 mm3; p = 0.039, Cohen d = 1.867), but not normalized (WT = 4.27; DMD = 4.02; p = 0.306) in brain volume. Diffusion tensor imaging (DTI) revealed structural changes in DMD temporalis muscle, with an increase in mean diffusivity (MD), axial diffusivity (AD), and radial diffusivity (RD).
In DMD mice thalamus, DTI revealed an increase in fractional anisotropy (FA) and a decrease RD. Smaller volume of normal brain correlated with the severity of muscular dystrophy (r = -0.975). neurochemical changes in the brains of mice DMD including increased GABA and NAA in the prefrontal cortex, and GABA in the hippocampus. The findings could indicate impaired motor and sensory signals, generating dysfunctional GABAergic neurotransmission and brain osmoregulasi unstable in dystrophin-zero.

In this study, after treating a healthy adult female mice with arsenite through drinking water for 6 months, our results clearly indicate that chronic exposure to arsenite not only disturbed gut microbiota composition but also cause neurobehavioral dysfunction, manifested with learning and memory deficits and dismay such behavior. Given that an overactive autophagy directly lead to intestinal pathological changes, we further assess whether inhibition of autophagy with average genetics can reverse the arsenite-induced neurobehavioral dysfunction.
Our results are illustrated for the first time that the heterozygous disruption of Beclin 1, which plays a central role in autophagy, alleviate intestinal disorders microbiome phenotype caused by arsenite, and ultimately lead to an increase in neurobehavioral deficits through the gut-brain communication. These findings provide new clues that regulation of autophagy is a potential approach to investigate the functional impact of arsenite on intestinal microbiome, and also can be cut as a way to strategy arsenite protection against neurotoxicity.
